Aging changes in cellular immune function in rats and Traditional Chinese Medicine anti-aging effect Tiannianyin
【Abstract】 Objective To observe the D-galactose aging rats
the changes in cellular immune function and anti-aging
medicine Tiannianyin role. Methods 40 SD rats were randomly divided into four groups, normal group, aging model group, their later years drinking drug group and negative control group were 10. D-galactose produced by intraperitoneal
injection of sub-acute rat model of aging. In each group using ELISA, serum IL-2 and IL-6 levels, while rats of the thymus detected index and phagocytic function of macrophages. The results of aging serum levels of IL-2, thymus index,
macrophage phagocytosis, compared with normal rats decreased
significantly (P <0.05, P <0.01), aging serum levels of IL-6
in normal significantly higher than in rats (P <0.01). Tiannianyin can significantly increase the aging rat serum IL-
2 content, the thymus index and phagocytic function of macrophages (drug in rats compared with the aging model group: P <0.01, P <0.05); Tiannianyin can significantly reduce the aging of serum IL-6 levels (drug in rats compared with the aging model group: P <0.01). Conclusion Tiannianyin can improve cellular immune function in aging rats, with a certain anti-aging effect.

Study on the Changes of Cellular Immune Function of Aging Rats and the Anti-aging Effects of Traditional Chinese
Abstract: ObjectiveTo study on the changes of cellular immune function of aging rats and the anti-aging effects of
traditional Chinese medicine Tiannianyin.Methods40 SD rats were divided into 4 groups at random: normal group, aging model group, Tiannianyin group and negative control group , each group had 10 rats. The sub-acute aging model rats were
made by injecting D-gal into abdominal cavity continually. ELISA was used to detect the IL-2 and IL-6 content in serum,
the macrophagic phagocytosis and thymus index were also
observed. ResultsCompared with normal rats, the IL-2 content
in serum, the thymus index and the macrophagic phagocytosis of aging rats decreased obviously (P <0.05, P <0.01); the IL-6
content in serum increased obviously (P < 0.01). Tiannianyin
not only significantly increased the IL-2 content, the
macrophagic phagocytosis and the thymus index of aging rats (Tiannianyin group compared with aging group: P <0.01, P <0.05); but also significantly decreased the IL-6 content of
aging rats (Tiannianyin group compared with aging group: P <0.01). ConclusionsTianninyin can strengthen the cellular immune function of aging rats, so it has fairly anti-aging

Studies have shown that with age, growth, immune function disorders, and the gradual decline occurred, due to a decline in immune function, resulting in autoimmune disease and cancer increase in . Traditional Chinese Medicine by Liu Hejian Tiannianyin "Vision on the side" Polygonum multiflorum

antioxidant capacity, improve learning and memory function and other aspects of the role of the [2 ~ 4], this paper observed Tiannianyin pairs of D-galactose aging rats The role of
cellular immune function, and further explore Tiannianyin anti-aging mechanism of action.
1 Materials and methods
1.1 Selection and grouping of animals selected
experimental SD rats were 40 male and female in half, weighing 180 ~ 250 g. The rats were randomly divided into four groups, normal group, aging model group, their later years drinking drug group and negative control group, 10 in each group. One model group, medication group, negative control rats are used D-galactose by intraperitoneal injection (at 200 mg / kg dose, 1 time / d, continuous 40 d) Production sub-acute animal model
of aging. Model group after modeling success will no longer do any processing; medication modeling in rats after the success of every day Tiannianyin gavage, a total of 30 d; negative control group, rats fed daily after the success of modeling
the same dose of saline, the time with the drug in rats.
1.2 Drugs and reagents Tiannianyin by the system, fleece-
flower root, Achyranthes bidentata, meat from Chengdu, epimedium, red sage root, Poria, etc. composed of six Chinese
herbal medicines, brewed with crude drug per ml after 1.5 g, according to 16.2 g / kg gavage.
D-galactose (D-gal) produced by Beijing Chemical Reagent Company (Lot No.: 020,402). Serum IL-2, IL-6 levels by ELISA
kit crystal US companies.
1.3 Index Detection
1.3.1 Serum IL-2 and IL-6 levels in rats were detected by
the inner canthal blood, taking the supernatant after centrifugation. Double-antibody sandwich ELISA, according to
kit instructions, were detected in serum IL-2 and IL-6 levels.
1.3.2 Detection of rat thymus index were decapitated, after weighing, remove the thymus, remove fat, fascia, after weighing, calculation of the thymus index: thymus index = thymus weight / body weight (mg/10 g).
1.3.3 macrophage function test according to literature determined by the method of macrophage phagocytosis, phagocytic index and phagocytic rate calculation.
1.4 Statistical analysis of all data to ? s said that
the use of SPSS 11.0 statistical analysis software to t test
was used for statistical processing.
2.1 Serum IL-2 and IL-6 content of the results in Table
1. Can be seen from Table 1, aging rats serum levels of IL-2
decreased significantly, IL-6 were significantly increased
(compared with normal rats: P <0.01); medicine Tiannianyin can significantly increase the serum IL-aging 2 levels, reducing
aging serum IL-6 levels, medication group compared with the model group significantly different (P <0.01). Table 1 rats in each group of serum IL-2 and IL-6 levels (omitted)
2.2 The index of rat thymus results in Table 2. Table 2 shows that the thymus index of aging rats compared with normal rats decreased significantly (P <0.01); Tiannianyin enhance the aging rat thymus index, treatment group compared with the
aging rats have a significant difference ( P <0.05).
2.3 in rat macrophage phagocytosis results in Table 2. Table 2 shows that aging rat macrophage phagocytosis decreased significantly (compared with normal rats: P <0.01, P <0.05);
their later years could significantly improve the aging rat macrophage phagocytosis (treatment group with the aging group: P <0.01, P <0.05). Table 2 rats in each group thymus index and the phagocytic function of macrophages (omitted) reposted

1977 Basedovsky for the first time that "immunity -
Nerve - endocrine network" (immune-neuroendocrine network)
doctrine; 1988 Meites proposed that in the aging process,
immune - nervous - endocrine network plays an important role; Many scholars study also shows that, due to immune function decline with age, resulting in autoimmune diseases and cancer has increased.
Although the body of the cell-mediated immunity and
humoral immune function in the growth of both change with age, but the decline in the elderly immune function primarily related to changes in T cells . And a study found that, T cell function to reduce degradation are closely linked with the thymus . With aging, when morphological and functional changes of the thymus, thymic epithelial cells, decreased expression of MHC molecules leading to reduction in the total number of T cells and antigen-specific T-cell immune function.
IL-2 mainly by activated T cells, in regulating the immune response, anti-tumor, etc. play an important role in immune function reflecting the strength of key indicators, IL-2
produced by reduction of the body can accelerate the aging process . IL-6 mainly by mononuclear macrophages, T
cells, B cells produce cytokines such as, not only in the immune system plays an important role, but also act on the nervous, endocrine systems, thus affecting the whole body of various functional systems activities. In aging and many age-
related diseases occur, IL-6 levels were elevated to varying
degrees; due to abnormal levels of IL-6 increase, could lead
to a series of immune function and endocrine dysfunction, and thus speed up the body's aging process .
